Reconstituting and Measuring Human Growth Hormone (Somatropin)
If you’ve just acquired a vial of lyophilized human growth hormone (somatropin), it’s essential to know how to properly reconstitute and measure it for safe and effective use.

Understanding the Dosage
The vial will indicate the amount of powder in either International Units (IU) or milligrams (mg). Generally, 1 mg is equivalent to about 3 IU. For our purposes, we’ll use the approximation of 1 mg = 3 IU, as this is commonly referenced.

Reconstitution Process
1. Prepare Your Materials: You’ll need bacteriostatic water (BW), sterile water, or liquid vitamin B12 for reconstitution. BW is preferred for longer storage, as it contains 0.9% benzyl alcohol, making it safe for up to three weeks.
2. Sanitize the Vials: Use an alcohol swab to clean the rubber stoppers of both the somatropin vial and the diluent vial.
3. Draw the Diluent: With a 3cc syringe and a 23 or 25 gauge needle, draw up your chosen diluent. The amount isn’t critical, but it should facilitate easy measurement later. For example:
o 1 ml per 10 IU vial means each 10 mark on a U100 insulin syringe equals 1 IU of somatropin.
4. Inject the Diluent: Angle the syringe to gently add the diluent along the side of the somatropin vial, avoiding direct contact with the powder.
5. Mix: After adding the diluent, gently swirl the vial to dissolve the powder completely. Do not shake it vigorously. Store the reconstituted somatropin in the refrigerator. If BW is used, it will be viable for three weeks; if sterile water, approximately five days.
Measuring Your Dosage
Once you’ve reconstituted the hormone, measuring your dosage accurately is crucial. Here’s how to do it:
1. Know Your Measurements: 1 ml = 1 cc = 100 IU.
2. Calculate Your Units: For instance, if you’ve added 1 cc of diluent to a 10 IU vial, you’ll have 100 IU total. Divide this by the number of IU in the vial:
o 100 IU / 10 IU = 10. This means every 10 mark on your U100 syringe equals 1 IU of somatropin.
3. Draw Your Dose: To draw 2 IU, simply pull the syringe to the 20 mark.

Final Tips
1. Keep track of the amount of somatropin you have and the diluent used.
2. Divide the diluent volume by the somatropin amount to know how much each unit corresponds to on the syringe.
3. Multiply the result by the number of units you wish to inject.
